  • Company develops multi-mechanism drugs for the treatment of cancer and inflammatory diseases.
  • Two of its product candidates are in Phase II clinical trials for cancer, with several others closely following in Phase I and pre-clinical development.
  • Because its products work through multiple mechanisms of action, greater economic potential may be possible due to its offering of several therapeutic applications.
  • Company has strong intellectual property rights, and owns or has licensed on an exclusive basis a total of 49 issued patents and patent applications in the US for its product candidates.

Company Overview

EntreMed, Inc. is a clinical-stage pharmaceutical company focused on developing next generation multi-mechanism drugs for use in oncology, a medical field that studies the diagnosis, treatment, and prevention of cancer. The company’s product candidates target both diseased cells and the blood vessels that nourish them. EntreMed is focused on developing drugs that are safe, convenient, and provide the potential for improved patient outcomes. The company’s goal is to develop and commercialize therapeutics based on its expertise in angiogenesis, cell cycle regulation, and inflammation. These are all processes vital to the progression of cancer and other diseases, and are the focus of the company’s three clinical product candidates. Clinical trials have enabled the company to identify new molecules and chemical entities that represent important targets in the treatment of cancer and inflammatory diseases.

The company is developing compounds that have broad therapeutic and commercial potential in oncology. To further advance its commercial objectives, the company will seek strategic alliances, licensing relationships, and co-development partnerships.

Product Candidates

The treatment of cancer is multifaceted, often requiring a combination of surgery, chemotherapy, radiotherapy, and palliative care. It has become increasingly clear to the scientific community that its treatment must match the complexity of this disease by attacking cancerous cells through a diversity of pathways. EntreMed’s compounds work through multiple mechanisms of action (MOA), meaning a single compound can attack a disease through multiple cellular pathways and even impact different diseases. The company developed its initial drug pipeline based on comprehensive research into the relationship between malignancy and angiogenesis (the growth of new blood vessels).

One of EntreMed’s lead drug candidates, Panzem® (2-methoxyestradiol or 2ME2) NCD, is currently being tested as a cancer therapeutic in five Phase 2 clinical trials and has been granted FDA designation in three cancer indications – including glioblastoma multiforme, multiple myeloma and ovarian cancer. Panzem® NCD has the potential to work through multiple mechanisms of action to attack cancer cells through multiple pathways that affect the formation and replication of tumor cells, and can interrupt the formation of blood vessels that nourish tumor cells and sustain tumor growth.

The company’s second lead drug candidate, MKC-1, is a novel cell cycle inhibitor currently in Phase 1 and 2 clinical trials for cancer. In May 2006, the company commenced clinical studies with its third clinical-stage compound, ENMD-1198, in patients with advanced cancer.

EntreMed is developing drug candidates that will be easy to use, with minimal interruption to the patient’s daily routine, as compared to other modes of drug administration. Its main focus is on oral drug delivery technologies and other administration routes.

Intellectual Property

All EntreMed pipeline programs are backed by strong intellectual property rights. Each product candidate is covered by issued or pending composition, method, and use patents. The company owns, or has licensed on an exclusive basis, a total of 49 issued patents and patent applications in the US for its product candidates. EntreMed has a total of 136 issued patents and pending patent applications in the US and other countries.

Economic Viability

The company’s current focus on the development of small molecule, multiple mechanism drug candidates may translate into lower product development, process development, and other associated costs when compared to biological products. Alternatively, greater economic potential may be possible because each of EntreMed’s compounds may offer several therapeutic applications.

Business Development

Although oncology is EntreMed’s principal clinical and commercial focus, recent data supports further development of its compounds in non-oncology applications as well (i.e. rheumatoid arthritis). The company will move forward, continuing to develop compounds for oncology and inflammatory diseases, while selectively exploring strategic alliances for its compounds in other therapeutic areas. EntreMed may also pursue codevelopment partners for its core pipeline product candidates to help accelerate their development and strengthen the development programs. In turn, the company can provide its codevelopment partners with substantial know-how relating to small molecules and peptides that inhibit processes like angiogenesis and inflammation.

Financial

As of December 31, 2006, EntreMed reported cash and short-term investments of approximately $50.6 million. Total expenses for 2006 were in line with its Phase II progress on Panzem® NCD and MKC-1, as well as its Phase I activity with ENMD-1198. The company has cash and short-term investments to support current and planned operations into the second half of 2008.
